A speech didn't kill JFK.
1. Old Joe Kennedy went to La Cosa Nostra (US mafia) for help during the 1960 campaign. When the JFK administration took over, however, RFK and DoJ went after LCN.
Further, LCN lost their casinos and hotels in Cuba because of Castro. The Disastrous Bay of Pigs invasion gave them a second reason to hate JFK.
